Hydroxysafflor Yellow Alleviates Ventricular Remodeling in Myocardial Infarction by Regulating the TGFβ1/Smads Signaling Pathway
Objective To investigate the effects of hydroxysafflor yellow on ventricular remodeling and the TGFβ1/Smads signaling pathway after myocardial infarction. Methods Myocardial infarction model was established by ligating the left anterior descending coronary artery, with sham surgery group as control.Rats were randomly divided into model group, low, medium and high dose hydroxysafflor yellow groups, and losartan group.Rats were dosed for 4 weeks.Changes of cardiac morphology were observed. The content of malonaldehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and catalase(CAT)in serum of rats in each group were analyzed. The expression of transforming growth factor(TGFβ1),SMAD2/3,and P-SMAD2/3 in myocardial tissue were detected by western blot. Results Compared with the sham-operated group, the model group increased cardiac volume, larger infarct area, concave and thinning left ventricular wall.Compared with the model group, the treatment groups exhibited no obvious concavity in the left ventricular wall, reduced infarct area and volume;decreased serum MDA levels, increased activities of SOD,GSH-Px,and CAT(P<0.05).Compared with the model group, the treatment groups decreased protein expression of TGFβ1 and P-SMAD2/3 in cardiac tissue(P<0.05).Conclusion Hydroxysafflor yellow could alleviate ventricular remodeling, relieve oxidative stress, inhibit the TGFβ1/Smads signaling pathway.
