Determination of Three Nucleoside Components in Fermentative Cordycepic Fungal Powder
Objective The content and cluster ratio of the three nucleosides(uridine,guanosine and adenosine)of fermentative cordycepic fungal powder were studied,aiming to provide reference for the quality standard improvement and the development of new products.Methods Using Waters 2489 high performance liquid chromatograph;column:Agilent ZORBAX SB-C18(4.6 mm×250 mm,5 μm);mobile phase:phase A was methanol,and phase B was 0.01 mol/L potassium dihydrogen phosphate solution;gradient elution;column temperature:25℃;detection wavelength:260 nm;flow rate:1.0 mL/min;sample quantity:10 μL.Results Uridine,guanosine,and adenosine had good linear relationships within the ranges of 0.127 5-50.989 6 μg/mL(r=0.999 9),0.111 1-44.459 5 μg/mL(r=0.999 9),and 0.135 1-54.055 3 μg/mL(r=0.999 9)respectively.The average recovery rates(n=6)were 97.89%(RSD=0.13%),103.69%(RSD=3.48%),and 95.92%(RSD=2.11%)respectively.Conclusion The method is simple to operate,stable and reliable,has high sensitivity and good specificity,and can be used to accurately detect the content of three nucleoside components in fermentative cordycepic fungal powder.
