Determination of Ceftazidime,Cefotaxime and Ceftriaxone in Human Serum and Cerebrospinal Fluid by HPLC
Objective A high performance liquid chromatography(HPLC)method for the determination of ceftazidime,ceftriaxone and cefotaxime concentrations in serum/cerebrospinal fluid(CSF)was developed and used in therapeutic drug monitoring.Methods The chromatographic column was Diamonsil C18(250 mm×4.6 mm,5.0 μm);methanol-0.01 mol/L potassium dihydrogen phosphate solution(formic acid adjusts pH to 3.0)was a mobile phase with gradient elution;the detection wavelength was 254 nm;the flow rate was 1.0 mL/min;column temperature was 25 ℃;ceftriaxone and cefotaxime were used as internal standards.The specificity,linearity,precision,stability and extraction recovery rate were investigated.Results The linearity of ceftazidime,cefotaxime and ceftriaxone in serum/CSF was good(r>0.999).The intra-day and inter-day precisions of serum/CSF were good(RSD<5%),and the recovery rates were within 90%-110%.It was stable(RSD<15%)at room temperature for 24 h,refrigerated for 48 h,repeated freezing-thawing for 3 times,6 h after sample processing and frozen at-20 ℃ for 50 days.Conclusion The HPLC method is simple,accurate,rapid and specific,and can be used to monitor drug concentration and prevent the occurrence of antibacteria-related encephalopathy.
