枳实提取物对功能性消化不良大鼠的促消化作用研究

目的 探讨枳实提取物对功能性消化不良(FD)大鼠的促消化作用。方法 大鼠随机分为总黄酮高、中、低剂量组,生物碱高、中、低剂量组,阳性对照组,模型对照组和空白对照组。除空白对照组外,其余组均造成FD模型,连续给予7天受试物后,测定胃残留率和小肠推进率,分离血清测定胃动素(MTL)、胃泌素(Gas)、血管活性肠肽(VIP)、5-羟色胺(5-HT)、5-羟色胺受体4(5-HTR4)含量。结果 给予总黄酮或高剂量生物碱后的FD大鼠小肠推进率均高于模型对照组(P<0。05);总黄酮组的胃残留率低于模型对照组(P<0。05)。给予总黄酮或高、中剂量生物碱后,FD大鼠VIP含量显著低于模型对照组(P<0。05);总黄酮组的MTL含量显著高于模型对照组(P<0。05)。另外,给予高、中剂量总黄酮后,FD大鼠的5-HT含量和5-HTR4含量均显著高于模型对照组(P<0。05)。结论 枳实生物碱能抑制FD大鼠的VIP分泌,从而促进小肠蠕动;而枳实总黄酮能够通过促进FD大鼠分泌MTL,抑制VIP分泌,改善FD大鼠的胃排空和小肠蠕动功能,还能促进5-HT分泌,加强其与小肠中的5-HTR4受体的结合来增强FD大鼠胃肠动力。
Study on the Digestive Promoting Effect of Fructus Aurantii Extract on Functional Dyspepsia Rats
Objective Explore the digestive promoting effect of fructus aurantii extract on functional dyspepsia rats.Methods Rats were randomly divided into high,medium,and low-dose groups of total flavonoids,high,medium,and low-dose groups of alkaloids,positive control group,model control group,and blank control group.Except for the blank control group,all other groups were subjected to FD models.After 7 consecutive days of administration of the test substance,the gastric residual rate and small intestine propulsion rate were measured,and serum levels of motilin(MTL),gastrin(Gas),vasoactive intestinal peptide(VIP),serotonin(5-HT),and serotonin receptor 4(5-HTR4)were measured by separating serum samples.Results The small intestine propulsion rate of FD rats after administration of total flavonoids or high dose alkaloids was higher than that of the model control group(P<0.05).The gastric residual rate of the total flavonoids group was lower than that of the model control group(P<0.05).After administration of total flavonoids or high and medium doses alkaloids,the VIP content was significantly lower than that of the model control group(P<0.05).The MTL content of the total flavonoids group was significantly higher than that of the model control group(P<0.05).In addition,after administration high and medium doses of total flavonoids,the 5-HT content and 5-HTR4 content in FD rats were significantly higher than those in the model control group(P<0.05).Conclusion Fructus aurantii alkaloids can inhibit VIP secretion in FD rats,thereby promoting small intestine peristalsis;the total flavonoids of fructus aurantii can improve gastric emptying and intestinal peristalsis in FD rats by promoting the secretion of MTL,inhibiting VIP secretion,and enhancing the secretion of 5-HT.It can also promote the secretion of 5-HT and strengthen its binding with 5-HTR4 receptors in the small intestine to enhance gastrointestinal motility in FD rats.
