Study on Extraction Technology and Quality Standard of Red Paste Lichong Granules
Objective:To optimize the extraction process of Hongjiang Lichong Granules and establish its quality stand-ard.Methods:Taking the content of geniposide as the evaluation index,the optimum extraction process of Hongjiang Lichong Granules was optimized by L9(34)orthogonal test design method.The thin layer chromatography(TLC)method was used to identify the stir-fried Gardenia jasminoides Ellis,vinegar Corydalis Rhizoma,Chuanxiong Rhizoma,Angelicae Sinensis Radix,Paeoniae Radix Alba,Paeoniae Radix Rubra and Moutan Cortex in the preparation.The con-tent of geniposide was determined by high performance liquid chromatography(HPLC).Results:The optimal extrac-tion process was as follows:the whole decoction pieces were decocted for 3 times,the first decoction was added with 9.5 times the total weight of the medicinal materials,and the second and third times were added with 8 times the amount of water,30 min each time.The TLC spots of stir-fried Gardenia jasminoides Ellis,vinegar Corydalis Rhizoma,Chuanx-iong Rhizoma and Angelicae Sinensis Radix were double negative,and the three negative TLC spots of Paeoniae Radix Alba,Paeoniae Radix Rubra and Moutan Cortex were clear,negative without interference,and good durability.The line-ar range of geniposide was 0.508 mg/mL-0.544 mg/mL,and the linear equation was y=0.065 4x-0.710 6,r2=0.999 9.The RSD of precision,repeatability and solution stability was less than 3％.The average recovery was 99.72％(n=6)and RSD was 2.34％.Conclusion:The extraction process of the preparation is feasible,and the estab-lished qualitative and quantitative method is convenient and reliable,which can be used as the quality control method of Hongjiang Lichong Granules.
