栉孔扇贝Tssk2基因的序列特征及表达分析

为研究栉孔扇贝(Chlamys farreri)睾丸特异性丝氨酸/苏氨酸激酶2基因Tssk2的序列和表达模式,利用荧光定量PCR及生物信息学分析研究了该基因的序列特征、各组织表达和性腺周期表达,结果显示该基因CDS为723 bp,编码240个氨基酸;进化树显示,栉孔扇贝Tssk2蛋白的进化地位与生物学分类地位一致;栉孔扇贝Tssk2具有Tssk家族的S_TKc保守结构域,含丝氨酸/苏氨酸蛋白激酶活性区域(Ⅵb区域)和ATP结合区,但ATP结合区域缺少Ⅰ子域.Tssk2在栉孔扇贝各组织和性腺发育周期中的表达显示,精巢是Tssk2表达量最高的组织,尤其是成熟期精巢中表达量最高,推测Tssk2在成熟精巢中发挥重要作用,或与精子的成熟相关.
Sequence Characteristics and Expression Analysis of Tssk2 Gene in Chlamys farreri
In order to study the sequence and expression pattern of testis-specific serine/threonine-protein kinase 2 gene(Tssk2)in Chlamys farreri,the sequence characteristics,tissue expression and gonadal cycle expression of Tssk2 gene are studied by qRT-PCR and bioinformatics analysis.The results are as follows:The CDS(coding se-quences)of Tssk2 is 723 bp long and encodes 240 amino acids.Phylogenetic analysis shows that the evolutionary status of Tssk2 protein in C.farreri is consistent with its biological classification status.Tssk2 possesses the S_TKc conserved domain of the TSSKs,including the serine/threonine protein kinase active region(Ⅵb region)and ATP-binding region,but the ATP-binding region lacks the Ⅰ subdomain.The expression of Tssk2 gene in various tissues and gonadal development cycles of C.farreri show that Tssk2 expression is the highest in the sperms,especially in the sperms at maturity,which shows that this gene may play a key role in mature testes and/or is relates to sperm maturation.
