Labdane Diterpenoids and Alkaloids from Fruits of Forsythia Suspensa Thunb.vahl and Their Anti-viral Activity
The chemical components in the anti-inflammatory and antiviral active parts and the antiviral activity of Forsythia suspensa are studied.The compounds in F.suspensa are separated and purified by silica gel column chro-matography,Sephadex LH-20 column chromatography and high performance preparation chromatography.The structures of the compounds are identified according to their physicochemical properties and spectral data,and the antiviral activity of some of the compounds is studied by MTT method.The results show that nine labdane di terpe-noids and six alkaloids are isolated from the chloroform and ethyl acetate parts of 95%ethanol extract of F.suspen-sa.The nine labdane diterpenoids are 19-hydroxy-ent-labda-8(17),13E-dien-15-oic acid(1),ent-labda-8(17),13Z-dien-15,19-dioic acid(2),ent-labda-8(17),13E-dien-15,19-dioic acid(3),3β-hydroxy-8(17),13E-lab-dadien-15-oic acid(4),enantio-labda-8(20),1 3E-dien-15,18-dioic acid(5),Forsyshiyanin A(6),Forsyshiya-nin B(7),18-hydroxy-7-oxolabda-8(9),13E-diene-15-oic aicd(8),and 3-hydroxy-8(17),13E-labdadien-15-oic acid(9).The six alkaloids are Forsyshiyanine A(10),(7R)-6,7-dihydro-7-methyl-5H-cyclopenta[c]pyridine(11),Forsyshiyanine B(12),(-)-bicuculline(13),(-)-egenine(14),and(-)-7'-O-methylegenine(15).Among these compounds,10 and 12 are discovered from F.suspensa for the first time,as well as from the plants of Forsythia genus.MTT results show that compounds 1,2,10 and 12 have inhibitory effects on the growth of H1N1 influenza virus and respiratory syncytial virus.
