藜麦愈伤组织诱导、增殖及组织细胞学观察

以藜麦Faro无菌种苗为试验材料,通过组织培养辅助细胞学观察建立愈伤组织诱导、增殖体系.试验结果表明,以子叶、胚轴及种胚为外植体均能诱导获得愈伤组织,但最佳培养基配方有明显差异:子叶外植体配方为MS+0.5 mg/L 2,4-D+0.5 mg/L 6-BA,胚轴外植体配方为MS+0.5 mg/L 2,4-D+0.05 mg/L 6-BA,而种胚外植体配方为MS+0.2 mg/L 2,4-D+0.5 mg/L ZT.优化筛选愈伤组织继代增殖培养基为MS+0.1 mg/L 2,4-D+0.2 mg/L 6-BA.增加蔗糖浓度有利于藜麦愈伤组织非胚性细胞向胚性细胞转化.
Establishment of Callus Induction and Proliferation System and Histological Observation of Chenopodium Quinoa Willd
The callus induction and proliferation system of quinoa Faro is established through tissue culture assisted cytological observation.The results show that the cotyledon,hypocotyl and seed embryo as explants can induce cal-lus,but there are significant differences in the optimal medium formula:cotyledon explants are MS+0.5mg/L 2,4-D+0.5mg/L 6-BA,cotyl explants are MS+0.5mg/L 2,4-D+0.05mg/L 6-BA,and seed embryo explants are MS+0.2mg/L 2,4-D+0.5mg/L ZT.Callus proliferation mediam is optimized for MS+0.1mg/L 2,4-D+0.2mg/L 6-BA.Increasing sucrose concentration is beneficial to the transformation of callus cells into embryogenic cells.
