新型HF-LPME-HPLC对蒙药荜茇生物碱含量测定研究

摘要
研究建立一种新型中空纤维膜液相微萃取联合高效液相色谱(HF-LPME-HPLC)测定蒙药荜茇中生物碱含量,对影响HF-LPME萃取效率的各项参数进行优化分析.中空纤维膜材料为偏氟乙烯纤维膜(空隙率>50%),正辛醇为萃取剂,萃取温度为室温,振荡速度为200 r/min,萃取时间为128 min.建立了新型HF-LPME的萃取方法,对蒙药荜茇中胡椒碱的R>5,与传统萃取方法相比,富集倍数为40倍.线性范围分别为0.006~0.01 mg/mL,相关系数(r)为0.999 9.相比于传统的萃取方法,该方法使用极少量的有机溶剂,可高效萃取并测定蒙药荜茇中生物碱的含量,打破了仅能在环境和食品中应用的限制,可以在各类化学基础实验中推广应用.
Abstract
A novel hollow fiber membrane liquid microextraction combined with high performance liquid chromatography(HF-LPME-HPLC)was established for the determination of alkaloids in Mongolian medicine pepper Piper longum L.,and the parameters affecting the extraction efficiency of HF-LPME were optimized and analyzed.The specific optimization scheme of the method is as follows:fiber membrane:structure hollow vinylidene fluoride,and the voids were>50%;extractant:n-octanol;temperature:room temperature;extraction mode:oscillating at the speed of 200 r/min;extraction time:128 min.The extraction method of HF-LPME was established.The R>5 and enrichment factor of piperine by HF-LPME-HPLC analysis method was 40.This method uses a very small amount of organic solvent compared to traditional extraction methods and can effectively extract and determine alkaloids in Mongolian medicine long pepper,which can be widely used in various basic experiments.
