Expression Levels of POT1,Dickkopf-1 and PTEN in Pituitary Adeno-mas with Different Clinical Features and their Correlation
To investigate the expression levels protection of telomeres 1(POT1),Dickkopf-1 and phosphatase and tensin homolog deleted on chromosome ten(PTEN)in pituitary adenomas with different clinical features and their correlation,the pathological tissue specimens of 54 patients with pituitary adenoma who underwent surgical treatment in the authors'hospital from August 2018 to December 2022 were retrospectively selected as the case group,and 63 normal pituitary tissue specimens collected at the same time were selected as the control group.The expression levels of POT1,Dickkopf-1,PTEN messenger ribonucleic acid(mRNA)between the case group and the control group were compared,the expression of POT1,Dickkopf-1,PTEN proteins in pituitary tumor tissues of patients with different clinical characteristics were compared,and the correlation of expression of POT1,Dickkopf-1 and PTEN in pituitary adenomas was analyzed by Pearson correlation.The expression level of POT1 mRNA in the the case group was higher than that in the control group(P<0.05),while the expression level of Dickkopf-1 and PTEN mRNA was lower than the control group(P<0.05).The positive rate of POT-1 protein of pituitary tumor tissue in patients with pituitary adenoma with high differentiation,stage Ⅲ~Ⅳ,tumor diameter ≥2 cm,invasion depth ≥6 cm,vascular space invasion and lymph node metastasis were higher than the patients with low differentiation,stage Ⅰ~Ⅱ,tumor diameter<2 cm,invasion depth<6 cm,no vascular space invasion and no lymph node metastasis(P<0.05),and the positive rate of Dickkopf-1 and PTEN protein of pituitary tumor tissue were lower than the patients with low differentiation,stage Ⅰ~Ⅱ,tumor diameter<2 cm,invasion depth<6 cm,no vascular space invasion and no lymph node metastasis(P<0.05).The results of Pearson correlation analysis showed that POT1 protein of pituitary tumor tissue was negatively correlated with Dickkopf-1 and PTEN protein(r=-0.713,-0.659,P<0.05).Dickkopf-1 protein was positively corre-lated with PTEN protein(r=0.547,P<0.01).POT1 was highly expressed in pituitary adenoma tissues,Dickkopf-1 and PTEN were low expressed in pituitary adenoma tissues.The expression of POT1,Dickkopf-1 and PTEN in pituitary adenoma tissues of patients with pituitary adenoma had certain correlation,which could participate in the disease progression.
