Relationship between Abnormal Expression of APP and Biological Behavior of Oral Squamous Cell Carcinoma
This study was aimed at investigating the relationship between abnormal expression of amyloid precursor protein(APP)and biological behaviors of oral squamous cell carcinoma(OSCC).The cultured human OSCC WSU-HN13 cells were divided into the uninfected cell group,the control plasmid group,the control shRNA group,the APP over expression group and the APP shRNA group through lipofection transfection.The expression of APP was detected using quantitative real-time polymerase chain reaction(qRT-PCR).The proliferative activity of cells at 24 h,48 h and 72 h after transfection was detected by CCK-8 assay.Cell migration ability was detected by cell wound scratch assay,cell invasion ability was detected by transwell cell invasion assay,and cell apoptosis rate was detected by flow cytometry.The relative expression level of APP mRNA in the APP over expression group was significantly higher than that in the uninfected cell group,the control plasmid group,the control shRNA group,and the APP shRNA group(P<0.05).The relative expression level of APP mRNA in the APP shRNA group was significantly lower than that in the uninfected cell group,the control plasmid group,and the control shRNA group(P<0.05).From 24 h to 72 h after transfection,cell proliferation activity of the APP over expression group was higher than that of the uninfected cell group,the control plasmid group,the control shRNA group and the APP shRNA group(P<0.05).From 48 h to 72 h after transfection,cell proliferation activity of the APP shRNA group was lower than that of the uninfected cell group,the control plasmid group and the control shRNA group(P<0.05).Cell migration ability and invasion cell count in the APP over expression group were higher than those in the uninfected cell group,the control plasmid group,the control shRNA group and the APP shRNA group(P<0.05).Cell migration ability and invasion cell count in the APP shRNA group were lower than those in the uninfected cell group,the control plasmid group and the control shRNA group(P<0.05).The apoptosis rate of the APP over expression group was lower than that of the uninfected cell group,the control plasmid group,the control shRNA group,and the APP shRNA group(P<0.05).The apoptosis rate of the APP shRNA group was higher than that of the uninfected cell group,the control plasmid group,and the control shRNA group(P<0.05).Abnormal expression of APP can promote the proliferation,migration and invasion of OSCC and inhibit cell apoptosis,which may serve as a new target for the diagnosis and treatment of OSCC.
