N-肉豆蔻酰基转移酶(NMT)存在于真菌和原生动物等真核生物中,N-肉豆蔻酰基化是一种由NMT催化的蛋白质脂质修饰和共翻译修饰过程,其在信号级联转导、膜泡运输和蛋白定位等细胞进程中发挥重要作用.研究证实,NMT对真菌、寄生虫、病毒和肿瘤细胞的存活至关重要,是一个具有前景的抗真菌、抗寄生虫、抗病毒和抗癌新靶点.综述以NMT为靶点的小分子抑制剂的研究进展,为开发新型NMT抑制剂提供参考.
Research Progress of N-myristoyltransferase Inhibitors
N-myristoyltransferase(NMT)is present in eukaryotes such as fungi and protozoa.N-myristoylation,a NMT-catalyzed process of protein lipid modification and co-translational modification,plays an important role in such cellular processes as signal cascade transduction,membrane vesicular transport,and protein localization.Research has confirmed the vital importance of NMT for the survival of fungi,parasites,viruses,and tumor cells,making it a promising new target for antifungal,antiparasitic,antiviral,and anticancer interventions.This review summarizes the research progress of small molecule inhibitors targeting NMT,aiming to provide some reference for the development of novel NMT inhibitors.
