基于网络药理学与分子对接探究宣肺振心方治疗射血分数保留型心力衰竭的作用机制

目的:基于网络药理学与分子对接探究宣肺振心方治疗射血分数保留型心力衰竭(HFpEF)的作用机制.方法:使用TCMSP数据库及已发表文献,检索宣肺振心方组成中药物黄芪、刺五加、红景天、丹参、川芎、防己、水红花子、延胡索的活性成分,利用SwissTargetPrediction平台进行有效成分靶点预测.通过GeneCards数据库及OMIM数据库检索HFpEF相关靶点,与中药有效成分相关靶点取交集,整合并运用Uniprot数据库进行基因校正.将交集靶点导入String中获取蛋白质互作网络PPI,并运用Cyoscape 3.7.0软件筛选核心靶点,构建"中药—成分—靶点"网络并进行拓扑分析.借助Metascape运用交集靶点进行基因本体GO功能富集分析及京都基因与基因组百科全书KEGG信号通路富集分析,筛选数据导入微生信网站进行可视化绘图.筛选核心靶点及对应活性成分进行分子对接处理.结果:通过TCMSP数据库及现有文献检索宣肺振心方组方中药活性成分,预测并去重后获得相关靶点390个.在Gene-Cards 和OMIM数据库搜索HFpEF并去重后获得疾病靶点2 053个,与中药活性成分靶点交集靶点167个.PPI网络分析并筛选出TNF、IL-6、1L-1β等7个核心靶点."中药—成分—靶点"网络分析显示槲皮素、山柰酚、草质素、β-谷甾醇等为核心活性成分.GO功能富集分析显示主要与蛋白激酶活性、G蛋白偶联胺受体活性、氧化还原酶活性等功能相关.KEGG通路富集分析筛选得到199条信号通路.结论:宣肺振心方中的槲皮素、山柰酚、草质素、β-谷甾醇等有效成分可能通过作用于TNF、IL-6、IL-1β等核心靶点,干预cGMP-PKG、钙、脂质和动脉粥样硬化等信号通路,从而对射血分数保留型心力衰竭患者起到干预心肌重构及改善心血管疾病预后的作用.
Study on Mechanism of Xuanfei Zhenxin Recipe in Treating Heart Failure with Preserved Ejection Fraction Based on Network Pharmacology and Molecular Docking Technique
Objective:This study was to explore the mechanisms of Xuanfei Zhenxin recipe in the treatment of heart failure with preserved ejection fraction(HFpEF)based on network pharmacology and molecular docking technique.Methods:We searched in TCMSP database and published literature for bioactive chemical components of Huangqi(Radix Astragali),Ciwu-jia(Acanthopanax Senticosus),Hongjingtian(Rhodiola RoseaL),Danshen(Salvia Miltiorrhiza),Chuanxiong(Ligusticum Chuanxiong Hort),Fangji(Radix Stephaniae Tetrandrae),Shuihonghuazi(Fructus Polygoni Orientalis)and Yanhusuo(Rhi-zoma Corydalis)in the composition of Xuanfei Zhenxin recipe.And then the effective component targets were predicted by SwissTargetPrediction platform.By searching for HFpEF-related targets in the Genecards and OMIM databases,intersection with the targets related to the effective components of Traditional Chinese Medicine in Xuanfei Zhenxin recipe.Subsequently,we integrated above targets and used the Uniprot database for gene corrections.The intersection targets were imported into String to obtain a protein-protein interaction network(PPI),and the Cyoscape 3.7.0 software was used to select core targets and construct a"Traditional Chinese Medicine-Component-Target"network for carrying out topological analysis.With the help of Metascape,the intersection targets were used for gene ontology(GO)functional enrichment analysis and Kyoto Encyclope-dia of Genes and Genomes(KEGG)signal pathway enrichment analysis,and then,selected data was imported into a bioinfor-matics website for visualization.Finally,core targets and corresponding active components were screened for molecular doc-king.Results:We searched bioactive chemical components of traditional Chinese medicine in Xuanfei Zhenxin recipe through TCMSP database and published literature after predicting and deduplicating,390 relevant targets were obtained.By searching the Genecards and OMIM databases for HFpEF and deduplicating,2 053 disease targets were obtained,and 167 intersecting targets with the active ingredient targets of traditional Chinese medicine were identified.It turned out that 7 core targets,in-cluding TNF,IL-6,and IL-1β,were analyzed and screened by PPI network.The"Traditional Chinese Medicine-Component-Target"network analysis showed that quercetin,kaempferol,luteolin,and β-sitosterol were core active ingredients.GO func-tion enrichment analysis showed that it was mainly related to protein kinase activity,G protein-coupled amine receptor activity and oxidoreductase activity.By enrichment analysis of KEGG pathway,199 signal pathways were obtained.Conclusion:The effective ingredients of Xuanfei Zhenxin recipe,such as quercetin,kaempferol,luteolin,and β-sitosterol,may be involved in the intervention of myocardial remodeling and the improvement of cardiovascular disease prognosis in patients with heart failure with preserved ejection fraction by acting on core targets such as TNF,IL-6,and IL-1 β and signaling pathways such as cGMP-PKG,calcium,lipids,and atherosclerosis.
