27羟基胆固醇在阿尔茨海默症中的作用机制

阿尔茨海默症(AD)是一种与年龄高度相关的疾病,遗传、其他代谢疾病等多种因素均参与其病理生理过程.迄今为止,AD的具体病理机制尚未明确,但对AD潜在病因的挖掘从未停止.最新的研究发现,高胆固醇血症加速了 AD的病理进程,外周胆固醇使生物大分子无法穿过血脑屏障,而在AD的致病分子中,血清胆固醇代谢物27羟基胆固醇可从外周流入大脑,在脑内参与AD的发生发展,因此靶向27羟基胆固醇可能是一种新的抗AD疗法.本文就27-HC与AD之间可能的作用机制进行综述,旨在为AD病理机制研究提供依据,从而为抗AD药物的研发开拓新思路.
The Mechanism of 27-hydroxycholesterol in Alzheimer's Disease
Alzheimer's disease is a highly age-related disease,and many factors such as genetics and other metabolic diseases are involved in its pathophysiological process.So far,the specific pathological mechanism of AD has not been clarified,but mining the potential etiology of AD has never stopped.Recent studies have found that hypercholesterolemia accelerates the pathological process of AD,and peripheral cholesterol is a biological macromolecular that cannot cross the blood-brain barrier.Among the pathogenic molecules of AD,serum cholesterol metabolite 27 hydroxycholesterol can flow into the brain from the periphery and participate in the occurrence and development of AD in the brain,so targeting 27-hydroxycholesterol may be a new anti-AD therapy.This article aims to review 27-hydroxycholesterol and its possible involvement in the process of AD,in order to provide a basis for the study of the pathological mechanism of AD,and to open up new ideas for the development of anti-AD drugs.
