Anti-inflammatory and Anti-oxidative Stress Effects of Epicatechin on Radiation-induced Stomatitis in Mice
Objective To investigate the protective effect of epicatechin on radiation stomatitis mice from the perspective of anti-inflammatory and anti-oxidative stress.Methods Thirty male C57 BL/6J mice were randomly divided into blank control group,model group and epicatechin group.The mouse model of stomatitis was established by single irradiation of 17 Gy X-ray.The epicatechin group was given epicatechin 30 mg/kg by gavage for 7 consecutive days after modeling,and the other two groups were given the same amount of normal saline by gavage.The oral inflammation score of mice in each group was evaluated.The pathological changes of oral mucosa were observed by HE staining.The levels of serum tumor necrosis factor-α(TNF-α),interleukin-1 β(IL-1 β)and interleukin-6(IL-6)were detected by enzyme-linked immunosorbent assay.And the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and catalase(CAT)and the level of malondialdehyde(MDA)in serum were detected by chemiluminescence.Results Compared with blank control group,the oral inflammation score,serum levels of TNF-α,IL-1β,IL-6 and MDA in model group were increased(P<0.05),while the serum activities of SOD,GSH-Px and CAT were decreased(P<0.05).Compared with model group,the oral inflammation score,serum levels of TNF-α,IL-1β,IL-6 and MDA in epicatechin group were decreased(P<0.05),while the serum activities of SOD,GSH-Px and CAT were increased(P<0.05).Conclusion Epicatechin has a good protective effect on mice with radiation stomatitis,and its mechanism may be related to the inhibition of inflammation and antioxidant damage.
