Network Pharmacological Analysis of the Mechanism of Furong Niaoganqing in the Treatment of Urinary Tract Infection
Objective To explore the potential targets,signaling pathways and mechanism of FurongNiaoganqing in the treatment of urinary tract infection based on network pharmacology.Methods The active components and targets of Furong Niaoganqing were obtained by TCMSP database and literature retrieval,and the gene name was corrected by Uniprot database.Genecards and DisGeNET databases were used to collect relevant targets for the treatment of urinary tract infection,and Venny2.1 was used to obtain drug-disease intersection targets.The protein-protein interaction(PPI)network of intersection targets was constructed by STRING database,and the topological analysis of core targets was carried out by Cytoscape software.GO and KEGG enrichment analysis of core targets was performed using DAVID database.Cytoscape3.9.1 software was used to construct a"drug-active ingredient-target-disease-pathway"network diagram.Results A total of 145 active components of Furong Niaoganqing were screened,mainly flavonoids such as quercetin,luteolin and baicalein,and 281 target genes were identified.A total of 10 308 disease targets and 262 intersection targets were collected.PPI protein network and topology analysis found key targets such as AKT1,TP53 and TNF;GO enrichment analysis showed that biological processes mainly involved gene expression,cell proliferation and apoptosis,inflammatory response,etc;cell components involve extracellular space,plasma membrane,nucleoplasm,etc.;the molecular functions involved ATP binding,protein kinase binding,DNA binding,etc.KEGG enrichment analysis obtained 147 pathways,mainly involving PI3K/Akt and Mark signaling pathways.Conclusion Furong Niaoganqing has the characteristics of multi-component,multi-pathway and multi-target in the treatment of urinary tract infection.Its mechanism may be to regulate PI3K/Akt and Mark signaling pathways by mediating the expression of key targets such as AKT1,TP53,TNF and IL6,so as to exert anti-inflammatory,inhibit bacterial activity and regulate body immunity,thereby eliminating urinary tract inflammation and improving clinical symptoms.
