Sinomenine Inhibits Migration and Invasion of Human Esophageal Cancer Cells by Regulating LncRNA-HEIH Expression
Objective To investigate the effect of sinomenine(SIN)on the migration and invasion of esophageal cancer cells and the regulation of LncRNA-HEIH.Methods Esophageal cancer TE-1 cells were treated with different concentrations of sinomenine.TE-1 cells were transfected with si-NC and si-HEIH,respectively.TE-1 cells were transfected with pcDNA and pcDNA-HEIH,respectively,and then treated with sinomenine.CCK8 was used to detect cell viability,scratch test was used to detect cell migration rate,Transwell test was used to detect the number of invasive cells,and qRT-PCR was used to detect HEIH expression.Results Sinomenine 25,50 and 100 mg/L had no significant effect on the viability of TE-1 cells(P>0.05).Under the action of sinomenine 25,50 and 100 mg/L,the migration rate and invasive cell number of TE-1 cells decreased significantly(P<0.01).Under the action of sinomenine 25,50 and 100 mg/L,the expression of HEIH in TE-1 cells decreased significantly(P<0.01).Transfection of si-HEIH significantly reduced the migration rate and the number of invasive cells of TE-1 cells(P<0.01).Overexpression of HEIH significantly reversed the effect of sinomenine on reducing the migration rate and the number of invasive cells of TE-1 cells(P<0.01).Conclusion Sinomenine inhibits the migration and invasion of esophageal cancer cells by inhibiting the expression of HEIH.
