摘要
目的 研究肝癌细胞NUAK家族激酶 2(NUAK2)对肝癌细胞侵袭和迁移能力的影响.方法 选取 2022 年 1 月-2023 年6 月在我院手术治疗的 40 例患者肝癌组织标本为观察组,并选取同期手术治疗存档的 40 例患者正常肝组织标本为对照组,采用聚合酶链式反应检测(PCR)技术和Western Blot 法检测两组NUAK2 及NUAK2 mRNA 的表达水平,Transwell 法检测NUAK2 对肝癌癌细胞株侵袭能力的影响,细胞划痕实验检测NUAK2 对肝癌癌细胞株迁移能力的影响.结果 观察组NUAK2水平、NUAK2 mRNA表达均高于对照组(P＜0.05);观察组肝癌细胞侵袭细胞个数、侵袭抑制率均低于对照组(P＜0.05);观察组肝癌细胞迁移细胞个数、迁移抑制率均低于对照组(P＜0.05).结论 NUAK2 在肝癌细胞组织呈现高表达水平,且会降低肝癌细胞侵袭、迁移率,进一步表明高水平NUAK2 与肝癌的侵袭和迁移能力密切相关,可能与其他蛋白相互作用后调控肝癌的发生与发展.
Abstract
Objective To study the effect of NUAK family kinase 2(NUAK2)on the invasion and migration of hepatocellular carcinoma cells.Methods From January 2022 to June 2023,40 cases of liver cancer tissue specimens from patients undergoing surgical treatment in our hospital were selected as the observation group,and 40 cases of normal liver tissue specimens from patients undergoing surgical treatment at the same time were selected as the control group.Polymerase chain reaction(PCR)and Western Blot were used to detect the expression levels of NUAK2 and NUAK2 mRNA in the two groups.Transwell method was used to detect the effect of NUAK2 on the invasion ability of liver cancer cell lines,and cell scratch test was used to detect the effect of NUAK2 on the migration ability of liver cancer cell lines.Results The level of NUAK2 and the expression of NUAK2 mRNA in the observation group were higher than those in the control group(P<0.05).The number of invasive cells and invasion inhibition rate of liver cancer cells in the observation group were lower than those in the control group(P<0.05).The number of migrated cells and migration inhibition rate of liver cancer cells in the observation group were lower than those in the control group(P<0.05).Conclusion NUAK2 is highly expressed in hepatocellular carcinoma tissues,and it can reduce the invasion and migration rate of hepatocellular carcinoma cells,which further indicates that high level of NUAK2 is closely related to the invasion and migration ability of hepatocellular carcinoma,and may regulate the occurrence and development of hepatocellular carcinoma after interacting with other proteins.
