Expression of signal transduction and activator of transcription 4 in ovarian cancer and its relationship with prognosis
Objective To investigate the expression of signal transducer and activator of transcription protein 4(STAT4)in ovarian cancer and its relationship with patient prognosis.Methods A total of 208 tissues were included in the tissue microarray,in-cluding 192 ovarian cancer tissues and 16 normal ovarian tissues.Immunohistochemistry was used to detect the expression of STAT4 in ovarian cancer tissues,paracancerous tissues and normal ovarian tissues.The clinicopathological characteristics and survival data of ovarian cancer patients were also obtained using the GEPIA database,the UALCAN database,the TCGA PORTAL database and the Kaplan-Meier Plotter database,which provided information on 419 ovarian cancer tissue samples from the GEPIA database and 1656 ovarian cancer tissue samples from the Kaplan-Meier Plotter data-base,which were used for analyzing the correlation between STAT4 expression in ovarian cancer tissues and tumour prognosis,respective-ly.Results STAT4 protein expression was significantly lower in ovarian cancer tissues(45.83%)compared to normal ovarian tissues(81.25%)(P<0.01).The GEPIA database showed that the overall survivalcurve(OS)of patients with STAT4 high-expression ovarian cancer was significantly higher than that of patients with low STAT4 expression(log-rank P=0.011),HR=0.73,P=0.012.Data from the TCGA Portal database showed that patients with high STAT4 expression had higher OS than patients with low STAT4 expression(log rank P=0.023).The Kaplan-Meier Plotter database re-sults showed that patients with high STAT4 expression had longer OS and progression-free survival(PFS)than patients with low STAT4 expression,and the difference was statistically significant(P=0.009,0.02).Conclusion STAT4 is significantly under-expressed in ovarian cancer tissues and is closely associated with patient survival and prognosis,and may be a potential indicator of ovarian cancer diagnosis and prognosis.
