Expression of miR-145-5p in serum of patients with essential hypertension and its impacts on the prolifera-tion and apoptosis of human umbilical vein endothelial cells
Objective To investigate the expression of miR-145-5p in the serum of patients with essential hypertension(EH)and its impacts on the proliferation and apoptosis of human umbilical vein endothelial cells(HUVEC).Methods From January 2019 to March 2022,160 patients with EH and 160 patients with normal physical examination results who were treated in Fenyang Col-lege of Shanxi Medical University were selected as the research objects.RT-qPCR was applied to detect the expression level of miR-145-5p in serum of EH patients and healthy subjects.HUVECs were cultured in vitro and divided into control group,miR-145-5p mim-ic group,mimic-NC group,miR-145-5p inhibitor group and inhibitor-NC group.The proliferation of HUVEC in each group was detected by CCK-8 method;AnnexinV-FITC/PI double staining was applied to de-tect the apoptosis of HUVEC in each group;Western Blot was applied to detect the expressions of HUVEC proliferation and apoptosis-related proteins[proliferating cell nuclear antigen(PCNA),Cyclin D1,B-cell lymphoma/leukemia gene-2-associated X protein(Bax),Caspase-3]in each group.Results The expression level of miR-145-5p in serum of EH patients was significantly lower than that of patients with normal physical examination results(P<0.05).Compared with the con-trol group and the mimic-NC group,the proliferation rate of HUVEC cells and the protein expressions of PCNA and Cyclin D1 in the miR-145-5p mimic group were increased,while the early apoptosis rate,late apoptosis rate and the protein expressions of Bax and Cas-pase-3 were decreased(P<0.05).Compared with the control group and the inhibitor-NC group,the proliferation rate of HUVEC cells and the protein expressions of PCNA and Cyclin D1 in the miR-145-5p inhibitor group were decreased(P<0.05),while the early apop-totic rate,late apoptotic rate,the protein expressions of Bax and Caspase-3 were increased(P<0.05).Conclusion miR-145-5p was lowly expressed in the serum of EH patients,and overexpression of miR-145-5p could promote the proliferation and inhibit the apopto-sis of HUVEC.
