The expression level of SUFU in oral squamous cell carcinoma and its effect on cell biological behavior
Objective To investigate the expression level of suppressor of fused(SUFU)in oral squamous cell carcinoma(OSCC)and the effect of low expression on the biological behavior of OSCC cells.Methods IHC was used to detect the expression of SUFU protein in OSCC tissues and adjacent normal tissues.The qRT-PCR and Western blot were used to measure the expression lev-els of SUFU mRNA and protein in OSCC cells(SCC15,CAL27)and human oral epithelial keratinocytes(HOK).Meanwhile,Pear-son Chi-square test was used to explore the relationship between histopprotein expression and clinicopathological features of OSCC pa-tients.The expression of SUFU was further reduced in OSCC cell lines,and the proliferation,invasion and migration ability of siSUFU and siNC cells were detected by CCK-8 proliferation assay,Transwell invasion assay and scratch healing assay.Results The expression level of SUFU protein in OSCC tissues was lower than that in adjacent normal tissues(P<0.05).The expression levels of SUFU mRNA and protein in OSCC cells were lower than those in human oral epithelial keratinocytes(P<0.05).The expression of SUFU protein was correlat-ed with the clinical stage and differentiation degree of OSCC patients(P<0.05).By transfecting SUFU interference fragments,it was found that the proliferation activity,the number of invasive cells and the scratch healing rate of siSUFU group were significantly increased compared with siNC group(P<0.05).Conclusion SUFU is low in OSCC,which is related to the clinical stage and differentiation of tumors,and is involved in regulating the proliferation,invasion and migration of OSCC cells,and may affect the occurrence and progression of OSCC by regulating Hh signaling pathway.
