O-GlcNAc糖基化修饰在肾脏疾病中的研究进展

O-GlcNAc糖基化(O连接N-乙酰氨基葡萄糖糖基化)修饰是常见的蛋白质翻译后修饰之一,涉及将O-连接的β-N-乙酰氨基葡萄糖(O-GlcNAc)添加在蛋白质上的丝氨酸/苏氨酸残基上,在各种细胞过程中起着重要作用,包括代谢、信号转导和细胞凋亡.O-GlcNAc糖基化修饰异常与多种疾病的发生密切相关,包括癌症、糖尿病、心血管疾病和神经退行性疾病等.近年来的研究表明,蛋白质O-GlcNAc糖基化修饰也广泛参与肾脏疾病的发生与发展.因此,为了解O-GlcNAc修饰在常见肾脏疾病中的变化,文章就O-GlcNAc糖基化修饰在肾脏(胚胎)发育、基因突变或肾脏发育异常相关性疾病中的研究进行综述,为未来继续研究肾脏疾病中O-GlcNAc糖基化修饰的扰动并以此为靶点研发出特异性治疗方式提供参考.
Research progress of O-GlcNAcylation modification in kidney diseases
O-GlcNAcylation(O-linked n-acetylglucosamine glycosylation)modification is one of the common protein post-translational modifications,involving the addition of O-linked β-n-acetylglucosamine(O-GlcNAc)to serine/threonine residues on pro-teins,and plays an important role in various cellular processes,including metabolism,signal transduction and apoptosis.Abnormal modification of O-GlcNAcylation is closely related to the occurrence of many diseases,including cancer,diabetes,cardiovascular dis-eases and neurodegenerative diseases.Recent studies have shown that the protein O-GlcNAcylation modification is also widely in-volved in the occurrence and development of kidney diseases.Therefore,in order to understand the changes of O-GlcNAc modification in common kidney diseases,this paper reviews the studies of O-GlcNAcylation modification in diseases related to kidney(embryonic)development,gene mutation or abnormal kidney development.It provides reference for further study of perturbation of O-GlcNAcyla-tion modification in kidney diseases and development of specific therapeutic methods based on this target.
