摘要

目的 检测猪内源性逆转录病毒(PERV)基因在异种肝肾联合移植供受体中的表达,以期为PERV的跨物种传播提供参考依据.方法 以6-基因编辑猪-恒河猴肝肾联合移植的组织样本作为研究对象,采用RT-PCR和PCR技术检测移植前后供受体不同组织中PERV编码基因、PERV各亚型以及猪线粒体细胞色素B基因的表达情况.结果 供体猪的不同组织中存在PERV编码基因gag和pol、PERV-A和PERV-B亚型以及猪线粒体细胞色素B基因,但不存在PERV-C亚型.移植终点时受体不同组织中均未检出PERV基因序列.结论 基因编辑猪-恒河猴异种肝肾联合移植受体存活的14 d内没有发生PERV的跨物种传播.

Abstract

Objective To detect the expression of porcine endogenous retrovirus(PERV)genes in donor and recipient of combined liver and kidney xenotransplantation,hoping to provide a reference for the cross-species transmission of PERV.Methods The tissue samples of 6-gene-edited pig-to-rhesus macaque combined liver and kidney xenotransplantation were selected as research objects.The expressions of PERV coding gene,PERV subtypes,and porcine mitochondrial cytochrome B gene in different tissues of donor and recipient samples during xenotransplantation were detected by RT-PCR and PCR.Results There were PERV coding genes gag and pol,PERV-A and PERV-B subtypes,and porcine mitochondrial cytochrome B gene in different tissues of donor pig,but no PERV-C subtype was detected in the donor tissue samples.At the end of transplantation,no PERV gene sequences were detected in different tissues of the recipient.Conclusion No cross-species transmission of PERV occurred within 14 d of survival in gene-edited pig-to-rhesus macaque combined liver and kidney xenotransplantation.
