摘要

异种实体器官移植是指将非人类来源的心脏、肾脏、肝脏等实体器官移植到人类受体中的一类探索性治疗技术.目前,来源于异种基因编辑猪肾脏、心脏的非人灵长类异种实体器官移植动物模型中已获得2~3年生存记录,因此,进一步改进基因编辑方式和效果,推进异种移植进入临床已成为器官移植的热点.2021-2023年,国外研究团队已经开展了十余例基因编辑猪-脑死亡遗体的肾脏和心脏移植亚临床研究,均未出现超急性排斥反应.本文综述近年报道的7例异种移植亚临床研究结果及2例猪心移植病例,讨论异种心脏、肾脏移植目前亟待解决的问题,为我国异种移植进入临床阶段研究提供一定的参考.

Abstract

Solid organ xenotransplantation refers to an exploratory therapeutic technique in which solid organs such as heart,kidney,and liver from non-human donors are transplanted into human recipients.Currently,survival records of 2 to 3 years have been obtained in animal models of solid organ xenotransplantation of non-human primates derived from heterogenous gene-edited pig kidneys and hearts.Consequently,further improving the way and effect of gene editing and advancing xenotransplantation to clinical application has become a hot spot in organ transplantation.From 2021 to 2023,international research teams have conducted more than 10 subclinical studies on kidney and heart transplantation of gene-edited pig-to-brain-dead decedents,none of which showed hyperacute rejection.This paper reviews the results of 7 studies on subclinical xenotransplantation and 2 cases of pig heart transplantation reported in recent years,and discusses the problems to be solved in heart and kidney xenotransplantation,so as to provide some references for the clinical research of xenotransplantation in China.
