摘要
目的 利用生物信息学分析系统阐述微管蛋白α-1B(TUBA1 B)与泛癌特征的关系.方法 利用TCGA数据库中的肿瘤患者临床资料,从表达水平、疾病分期、预后、基因突变特征、免疫浸润水平和基因富集分析6 个方面探讨 TUBA1 B 与各种肿瘤类型的相关性.结果 TUBA1 B 的表达水平在多数肿瘤中高于癌旁(P<0.05),在包括侵袭性乳腺癌等14 种肿瘤中具有早期诊断价值(P<0.05),在肾上腺皮质癌等3 种肾脏肿瘤中具有晚期进展监测价值(P<0.05),且高表达TUBA1 B在多种肿瘤预示较差预后(P<0.05).免疫浸润分析提示TUBA1 B与泛癌中多数免疫浸润细胞及多种免疫相关基因表达呈正相关(P<0.05).基因富集分析表明TUBA1 B可能通过调控细胞周期、DNA损伤修复、有丝分裂等生物学过程参与肿瘤发生发展.结论 TUBA1 B参与了多种肿瘤的发生发展和免疫浸润,可作为泛癌的肿瘤标志物和潜在的药物靶点.
Abstract
Objective To illustrate the relationship between tubulin alpha-1B(TUBA1 B)and pan-cancer features through bioinformatics analysis.Methods Clinical data of cancer patients in TCGA database were used to explore the correlation between TUBA1 B and various cancer types from six aspects:expression levels,disease staging,prognosis,gene mutation characteristics,immune infiltration level and gene enrichment analysis.Results TUBA1 B was highly expressed in most cancers compared with adjacent non-tumor tissues(P<0.05).TUBA1 B showed early diagnostic value in 14 tumors,including invasive breast cancer(P<0.05),and late progression monitoring value in 3 renal tumors,including adrenocortical carcinoma(P<0.05).High expression of TUBA1B indicated a poor prognosis in multiple tumors(P<0.05).Immunoinfiltration analysis revealed that TUBA1 B was positively correlated with most immune-infiltrating cells and immune-related genes in pan-cancer(P<0.05).Gene enrichment analysis indicated that TUBA1 B may be involved in the development of tumor by regulating biological processes such as cell cycle,DNA damage repair,and mitosis.Conclusion TUBA1 B is involved in the occurrence,development and immunoinfiltration of a variety of tumors,which can be used as a tumor marker for pan-cancer and a potential drug target.
