Objective To investigate the effect of IL-2/anti-IL-2 antibody on traumatic brain injury(TBI)in mice by increasing the number of regulatory T cells(Treg)in the central nervous system.Methods Healthy male C57BL/6 mice aged 10 weeks were divided into Sham group,TBI group and TBI+IL-2/anti-IL-2 antibody group.The TBI+IL-2/anti-IL-2 antibody group was intraperitoneally injected with IL-2(1.0 μg)and anti-IL-2 antibody(5.0 μg)for 3 consecutive days at 2 h after modeling.Flow cytometry was performed on the brain tissues of the three groups of mice at 3,5 and 7 d to detect the number of Treg.Immunofluorescence staining was performed on the brain tissues of mice on the 7th day to determine the presence of Treg infiltration in the central nervous system after TBI.In addition,the behavioral results of the three groups of mice at 1,3,5,and 7 d were analyzed,respectively.At the same time,the water content of brain tissues,neuronal apoptosis and the expression of inflammatory factors were detected on the 7th day of modeling.Results Compared with Sham group,there was Treg infiltration in the central nervous system of mice in TBI group and TBI+IL-2/anti-IL-2 antibody group,and the number of Treg gradually increased after TBI.There was no Treg infiltration in the Sham group.Compared with TBI group,TBI+IL-2/anti-IL-2 antibody group had a significant increase in Treg(P<0.05),including a reduction in neuronal apoptosis,brain edema,and expression of inflammatory factors,and a significant improvement in prognosis.Conclusion IL-2/anti-IL-2 antibody can increase the number of Treg in the central nervous system and alleviate TBI symptoms in mice to a certain extent.
