摘要

目的 通过比较甘肃省武威市边缘革蜱核糖体各片段的基因差异,确定边缘革蜱合适的分子鉴定方法.方法 2022年3月采集武威市边缘革蜱128只,随机选择10只进行实验.首先使用体式显微镜对蜱进行形态学鉴定,然后扩增边缘革蜱的5.8S、18S、28S、ITS1和ITS2核糖体序列并测序比对.使用MEGA 11软件构建蜱种的系统发育树,再通过DNASTAR软件比对边缘革蜱与长角血蜱等常见蜱种的基因同源性,进行基因多态性的分析和有效标记的筛选.结果 形态学鉴定结果显示所有蜱为同一种蜱,均为边缘革蜱.系统发育关系显示,核糖体18S、28S、ITS1和ITS2序列可区分革蜱属的蜱虫,5.8S序列无区分能力.同时18S和ITS1序列能将边缘革蜱与其他蜱种区分开,而且18S序列还可区分不同地域的边缘革蜱.结论 核糖体18S序列可区分边缘革蜱不同的地域株,其作为鉴定边缘革蜱分子学依据的能力较强.

Abstract

Objective To determine an appropriate molecular identification method for Dermacentor marginatus by comparing the gene differences in ribosomal fragments of Dermacentor marginatus in Wuwei City,Gansu Province.Methods In March 2022,128 Dermacentor marginatus were collected from Wuwei City,and 10 were randomly selected for the experiment.First,morphological identification of ticks was performed by a stereomicroscope.Then,ribosome sequences of 5.8S,18S,28S,ITS1 and ITS2 of Dermacentor marginatus were amplified and sequenced.MEGA 11 software was used to construct the phylogenetic tree of tick species,and then DNASTAR software was used to compare the genetic homology of Dermacentor marginatus and other common ticks such as Haemaphysalis longicornis.Results The results of morphological identification showed that all ticks were the same species—Dermacentor marginatus.Phylogenetic relationships showed that ribosome 18S,28S,ITS1 and ITS2 sequences could distinguish ticks of genus Dermacentor from other different species,while 5.8S sequence had no distinguishing ability.Meanwhile,18S and ITS1 sequences could distinguish Dermacentor marginatus from other ticks,and 18S sequence could also distinguish Dermacentor marginatus from different regions.Conclusion The ribosome 18S sequence can distinguish different geographical strains of Dermacentor marginatus and has a strong ability as an effective marker for molecular identification of Dermacentor marginatus.
