基于HPCPC白首乌C21甾体苷诱导PC-3细胞凋亡研究

目的 从白首乌块根中提取纯化C21甾体苷类化合物,并研究其诱导PC-3肿瘤细胞凋亡作用.方法 运用高效离心分配色谱,从白首乌总苷中分离纯化C21甾体苷,并利用FCM检测对PC-3细胞周期和凋亡的影响,利用AO荧光染色观察凋亡细胞形态学.结果 提取物对PC-3细胞的诱导凋亡作用具有量-效关系,且可以浓度依赖性的提高细胞G2/M期比例;AO染色荧光显微镜观察,提取物可诱发明显的核断裂和核固缩,以及凋亡小体的形成.结论 运用高效离心分配色谱分离纯化白首乌中的C21甾体苷,提取物能诱导PC-3肿瘤细胞凋亡,发生G2/M期阻滞,这可能是它抗肿瘤作用机制之一.
Studies on PC-3 cell apoptosis-inducing effect of C21 steroidal glycosides from Radix Cynanchi Auriculati by high performance centrifugal partition chromatography
Objective To extract and purify of C21 steroidal glycosides from Radix Cynanchi Auriculati and to study their apoptosis-inducing effects on PC-3 tumor cells. Methods By using high performance centrifugal partition chromatography, isolate and purify of C21 steroidal glycosides from the crude steroids of Radix Cynanchi Auriculati.The extract was tested by FCM and acridine orange (AO) staining fluorescence to study the apoptosis-inducing effect. Results FCM assay showed that the apoptosis-inducing effect was dose-depend with the extract, and the proportions of G2 cells and M cells increased; the results of AO staining fluorescence indicated that the extract induced nuclear fragmentation, pycnosis, formation of apoptotic body. Conclusion By using HPCPC, isolate and purify of C21 steroidal glycosides from Radix Cynanchi Auriculati, and the extract could induce apoptosis on PC-3 tumor cell and the arrest of cell cycle G2/M phase, which may be the one of mechanisms of anti-tumor effect against PC-3 cells.
