蒌蒿抗肿瘤作用活性部位的筛选

Screening of antitumor active fractions from Artemisia Selengensis Turcz.

目的:采用体内外抗肿瘤实验方法,筛选蒌蒿抗肿瘤活性部位。方法以95%乙醇提取,依次用石油醚、氯仿、乙酸乙酯、水饱和正丁醇提取,得不同的提取层。用四甲基偶氮唑蓝(MTT)法筛选出对人肝癌细胞(HepG2)细胞有良好增殖抑制作用的活性部位,并通过荷瘤小鼠模型考察活性部位对实体瘤生长的影响。结果蒌蒿的乙酸乙酯层对细胞增殖有显著的抑制作用,在10 g/L浓度时对细胞增殖抑制率为15.64%,当浓度为1000 g/L时对细胞增殖抑制率高达78.21%,其作用随着质量浓度的增加而增强。蒌蒿的乙酸乙酯层也对荷瘤小鼠有显著的抑制作用,当浓度为400 mg/kg时,对小鼠HepG2肉瘤的抑瘤率达71.63%。结论蒌蒿的乙酸乙酯层具有显著的体内、外抗肿瘤活性。
Objective To screen the antitumor active fraction from Artemisia selengensis Turcz. by antitumor experi-mental method in vitro and in vivo. Methods Artemisia selengensis Turcz. were extracted with 95% ethanol eluent, then petroleum ether, chloroform, ethyl acetate, n-butyl alcohol extracted from the active fraction. The active fraction of good inhibitory effect on HepG2 cells proliferation was screened out by MTT assay. The inhibition effect of solid tumor by tumor-bearing mouse model was investigated. Results Ethyl acetate extract of Artemisia selengensis Turcz. could significantly inhibit the growth for cell proliferation, the inhibition rate was 15.64% at 10 g/L. The inhibition rate was 78.21% at 1000 g/L. With the increase of the concentration, inhibiting action increased. Ethyl acetate extract of Artemisia selengensis Turcz. significant inhibition effect of solid tumor. When the concentration of 400 mg/kg for tumor bearing mouse inhibition rate was 71.63%. Conclusion Ethyl acetate extract of Artemisia selengensis Turcz. has signif-icant anti-tumor activity in v itro and in v iv o.
