Expression of signal transducer and activator of transcription 3 in clear cell renal cell carcinoma and its relationship with clinicopathological features and immune infiltration
Objective To investigate the expression of signal transducer and activator of transcription 3(STAT3)in clear cell renal cell carcinoma(ccRCC)and its correlation with clinicopathological features and immune infiltration.Methods From March 2017 to January 2022,cancer tissue samples of 105 ccRCC patients and 43 paracancer tissue samples were collected from the Department of Urology,General Hospital of the Eastern Theater Command of the Chinese People's Liberation Army.Hematoxylin-eosin staining,tissue microarray(TMA),and immunohistochemical(IHC)staining were used to detect STAT3 expression in the tissues.According to their staining scores,ccRCC patients were divided into high STAT3 expression(≥two points)and low STAT3 expression patients(<two points),and the relationship between its expression and clinicopathological characteristics was analyzed.The association between STAT3 gene expression and ccRCC immune infiltration was analyzed by TIMER and TISDB databases.Results Hematoxylin-eosin staining showed that immune cells were dispersed in ccRCC tissue.TMA-IHC analysis showed that the positive expression rate of STAT3 protein in ccRCC cancer tissues was higher than that in paracancer tissues,the differences were statistically significant(P<0.05).STAT3 expression was correlated with tumor size,Fuhrman grade,T stage,and lymph node metastasis in ccRCC patients(P<0.05).The analysis of TIMER database showed that STAT3 expression was positively correlated with the infiltration levels of B cells,CD8+T cells,CD4+T cells,macrophages,neutrophils,and dendritic cells(partial correlation coefficient>0,P<0.05).TISIDB database analysis showed that STAT3 expression was positively correlated with the abundance of natural killer cells and regulatory T cells(rs>0,P<0.05),and negatively correlated with the abundance of CD56bright and CD56dimnatural killer cells(rs<0,P<0.05).Conclusion The high expression of STAT3 is related to the clinicopathological features of ccRCC,and STAT3 is related to tumor immune infiltration,which is of great significance in regulating the occurrence and development of ccRCC.
