高效液相色谱法指纹图谱结合化学模式识别对除湿丸质量控制研究

目的 建立除湿丸指纹图谱,为完善其质量控制提供参考.方法 采用高效液相色谱法(HPLC),色谱柱为Luna Omega Polar C18 色谱柱(150 mm×4.6 mm,5 μm),流动相:乙腈-0.2%磷酸水溶液,流速为 1.0 ml/min,波长为254 nm,柱温30℃,进样量 10 μl.以黄芩素为参照,测定 29批除湿丸的HPLC图谱,并采用中药色谱指纹图谱相似度评价系统(2012 版)进行相似度评价.结果 29 批除湿丸HPLC指纹图谱有 35 个共有峰,相似度均>0.95,并指认了 8 个色谱峰;聚类分析将 29 批除湿丸聚为 3 类;主成分分析中 7 个主要成分分别反映 32 个共有峰信息.结论 不同批次除湿丸存在一定的质量差异.所建立的除湿丸指纹图谱可全面评价除湿丸的质量.
Study on quality control of Chushi Wan by high performance liquid chromatography fingerprint combined with chemical pattern recognition
Objective To establish the fingerprint of Chushi Wan and to provide the reference for improving the quality control.Methods Luna Omega Polar C18 column(150 mm×4.6 mm,5 μm)was used in this high performance liquid chromatography(HPLC),mobile phase:acetonitrile-0.2%phosphoric acid aqueous solution,flow rate was 1.0 ml/min,wavelength was 254 nm,column temperature was 30℃,injection volume was 10 μl.The HPLC chromatograms of 29 batches of Chushi Wan were determinated and evaluated by the similarity evaluation system of traditional Chinese medicine chromatographic fingerprint(2012 edition)with the reference of baicalein.Results There were 35 common peaks in the HPLC chromatograms of 29 batches of samples,the similarity was above 0.95 and eight chromatographic peaks were identified.Cluster analysis grouped 29 batches of Chushi Wan into three groups.The seven main components in principal component analysis reflected 32 common peaks information.Conclusion There are some quality differences in different batches of Chushi Wan.The established fingerprint can be used to evaluate the quality of Chushi Wan.
