Relationship between the expression of microRNA-296,vascular endothe-lial growth factor-B and the degree of coronary artery disease in patients with coronary heart disease
Objective To investigate relationship between the expression of microRNA-296(miR-296),vascular endothelial growth factor-B(VEGF-B)and the degree of coronary artery disease in patients with coronary heart disease(CHD).Methods A total of 93 patients with CHD admitted to the Second People's Hospital of Liaocheng from January 2019 to November 2022 were selected.They were divided into single vessel lesion group of 38 cases,dual vessel lesion group of 33 cases,and multi vessel lesion group of 22 cases based on the number of coronary artery lesions.According to the Gensini score,they were divided into coronary artery stenosis(CAS)mild group of 30 cases,moderate group of 34 cases,and severe group of 29 cases.Real-time fluorescence quantitative polymerase chain reaction was used to detect miR-296,and enzyme linked immunosorbent assay was used to detect serum VEGF-B.The relationship between miR-296 and VEGF-B was verified using dual luciferase reporter genes.Pearson correlation was used to analyze the correlation between miR-296 and VEGF-B expression in CHD patients,and Spearman correlation was used to analyze the correlation between miR-296 and VEGF-B expression,Gensini score,and the number of coronary lesions in CHD patients.Results The expression of miR-296 in the multi vessel lesion group was lower than that in the single vessel lesion group and dual vessel lesion group,and the level of VEGF-B was higher than that in the single vessel lesion group and dual vessel lesion group,the expression of miR-296 in the dual vessel lesion group was lower than that in the single vessel lesion group,and VEGF-B was higher than that in the single vessel lesion group,and the differences were statistically significant(P<0.05).The expression of miR-296 in the severe group was lower than that in the mild and moderate groups,and the level of VEGF-B was higher than that in the mild and moderate groups,the expression of miR-296 in the moderate group was lower than that in the mild group,and the level of VEGF-B was higher than that in the mild group,and the differences were statistically significant(P<0.05).The dual luciferase reporter genes showed that VEGF-B was the target of miR-296.Pearson correlation analysis showed a negative correlation between miR-296 and VEGF-B expression in CHD patients(r=-0.731,P<0.05),Spearman correlation analysis showed a negative correlation between miR-296 expression and Gensini score and lesion number in CHD patients(rs=-0.719,0.638,P<0.05),and a positive correlation between VEGF-B expression and Gensini score and lesion number(rs=0.727,0.698,P<0.05).Conclusion The low expression of blood miR-296 and high expression of VEGF-B in CHD patients are closely related to the degree of CAS lesions.
