Distribution of salidroside in Lujiao Fang in the myocardial tissues of rats with chronic heart failure based on LC-MS/MS
Objective To analyze the distribution of salidroside in Lujiao Fang in myocardial tissues of rats with chronic heart failure.Methods A total of 45 SPF male Wistar rats,aged 6-8 weeks,were selected.A simple random sampling method was used to select eight rats as sham operation group.In sham operation group,only the abdominal aorta was exposed without ligature;the left rats underwent abdominal aortic coarctation,and the obvious coarctation at the vessel ligation site indicated successful operation.After operation,according to the body weight of 32 surviving rats,model rats were randomly divided into the model group(n=8),low-dose Lujiao Fang group(1.100 g/kg,n=8),medium-dose Lujiao Fang group(2.200 g/kg,n=8),and high-dose Lujiao Fang group(4.400 g/kg,n=8),and given intragastric intervention of drug for 30 weeks.The rats were anesthetized,blood was collected from the abdominal aorta,and the hearts were collected 2 h after the last intragastric administration.LC-MS/MS method was used to analyze the distribution of salidroside in left and right ventricular wall of rats with different doses.Results Salidroside had a good linear relationship in the range of 1.195-1 195.000 ng/ml(r＞0.999),precision RSD％＜15％,accuracy was 90.817％-106.500％,relative recovery was 95.222％-100.814％,substrate effect RSD％＜15％.The stability of salidroside was good after the sample was placed in ice bath for 1 h,at room temperature for 6 h after pretreatment,and 48 h after automatic sampler.The linear fitting value R2of salidroside content in right ventricular wall and dose was 0.983.The content of salidroside in left ventricular wall was(47.383± 12.156)％and(43.805±9.644)％of right ventricular wall,respectively.Conclusion The established analytical method is feasible.The distribution of salidroside in left and right ventricular wall of chronic heart failure rats is different.
