Study on the mechanism of Jianpi Jiedu Decoction in the treatment of pso-riasis rats with syndrome of dampness stagnancy due to spleen deficiency based on oxidative stress and inflammation
Objective To investigate the mechanism of Jianpi Jiedu Decoction in the treatment of psoriasis rats with syndrome of dampness stagnancy due to spleen deficiency based on oxidative stress and inflammation.Methods A total of 48 SPF grade SD male rats aged 6-8 weeks and weighing 180-200 g were divided into blank group,model group,positive drug control group,and Jianpi Jiedu Decoction low-dose,medium-dose,and high-dose groups according to random number table method,with eight rats in each group.After hair removal,Vaseline was applied to the back of blank group,and other groups were smeared with 5% Imiquimod Cream(62.5 mg,twice a day)and intragastric administration Radixet Rhizoma Rhei Granules(10 ml/kg,twice a day),and the model of psoriasis with syndrome of dampness stagnancy due to spleen deficiency was established for ten days.After successful modeling,the Jianpi Jiedu Decoction low-dose,medium-dose,and high-dose groups were given 2.32,4.64,and 9.26 g/kg Jianpi Jiedu Decoction,respectively.The positive control group was given 0.97 mg/kg Methotrexate Tablets by intragastric administration;blank group and model group were given the same amount of normal saline.After 14 days of administration,psoriasis area and severity index(PASI),histopathology,contents of interleukin-22(IL-22),malondialdehyde(MDA),glutathione(GSH),activity of superoxide dismutase(SOD),and expression of heme oxygenase-1(HO-1)protein were compared among the six groups.Results The erythema,scale,infiltration,feeding,and defecation of Jianpi Jiedu Decoction low-dose,medium-dose,and high-dose groups were better than those in model group.PASI in Jianpi Jiedu Decoction low-dose,medium-dose,and high-dose groups were lower than those in model group,PASI in Jianpi Jiedu high-dose group was lower than that in Jianpi Jiedu Decoction low-dose,medium-dose groups(P<0.05).Compared with the model group,the epidermal hyperplasia and hyperkeratosis of Jianpi Jiedu Decoction low-dose,medium-dose,and high-dose groups were reduced,epidermal mutation was shorter,and dermal vasodilation was weakened.SOD activity,GSH and HO-1 levels in model group were lower than those in blank group,MDA and IL-22 levels were higher than those in blank group(P<0.05).The serum levels of GSH in Jianpi Jiedu Decoction low-dose,medium-dose,and high-dose groups were higher than those in model group,serum SOD activity in Jianpi Jiedu Decoction medium-dose,high-dose groups were higher than those in model group,serum MDA and IL-22 were lower than those in model group,and skin tissue HO-1 in Jianpi Jiedu Decoction high-dose group was higher than that in model group(P<0.05).Serum SOD activity and GSH level in Jianpi Jiedu Decoction medium-dose and high-dose groups were higher than those in Jianpi Jiedu Decoction low-dose group,serum MDA and IL-22 levels were lower than those in Jianpi Jiedu Decoction low-dose group,and skin tissue HO-1 in Jianpi Jiedu Decoction high-dose group was higher than that in Jianpi Jiedu Decoction low-dose group(P<0.05).The serum SOD activity,GSH,and skin tissue HO-1 levels in Jianpi Jiedu Decoction high-dose group were higher than those in Jianpi Jiedu Decoction medium-dose group,and serum MDA was lower than those in Jianpi Jiedu Decoction medium-dose group(P<0.05).Conclusion Jianpi Jiedu Decoction can improve the pathological symptoms of psoriasis rats with syndrome of dampness stagnancy due to spleen deficiency,and it is related to reducing the inflammatory index in serum,playing the role of anti-infection and anti-oxidative stress.
