Study of the relationship between the expression of miR-525-5p,perilipin 3 and the prognosis of glioma
Objective To investigate the expression of micro ribonucleic acid-525-5p(miR-525-5p)and perilipin 3(PLIN3)in glioma tissues and their relationship with prognosis.Methods A total of 102 glioma patients admitted to Qingdao Municipal Hospital from February 2018 to July 2020 were selected,and some intraoperative tumour tissues and paratumour tissues were collected,and miR-525-5p and PLIN3 mRNA expression were detected by real-time fluorescence quantitative polymerase chain reaction,and PLIN3 was detected by immunohistochemistry.The binding sites of miR-525-5p and PLIN3 were predicted by TargetScan database,and the correlation of their expression in glioma tissues was analysed.According to the mean value of PLIN3 mRNA expression in glioma tissues were divided into high-expression group and low-expression group,Kaplan-Meier method was used to draw the survival curves of glioma patients with high/low PLIN3 mRNA expression,and multifactorial Cox regression was used to analyse the factors affecting the prognosis of glioma patients.Results Compared with paraneoplastic tissues,miR-525-5p expression was reduced and PLIN3 mRNA expression was elevated in glioma tissues(P<0.05).Compared with paraneoplastic tissues,PLIN3 protein positive expression was elevated in glioma tissues(P<0.05).There was a binding site for miR-525-5p and PLIN3.miR-525-5p was negatively correlated with PLIN3 mRNA expression in glioma tissues(P<0.05).PLIN3 mRNA high expression had a lower 3-year overall survival rate than the lower expression group(P<0.05).Multivariate Cox regression analysis showed that low differentiation,World Health Organization Central nervous system tumors gradeⅢ-Ⅳ,and PLIN3 mRNA≥1.86 were independent risk factors for death in glioma patients(P<0.05).Conclusion The expression of miR-525-5p is low in glioma tissues,while the expression of PLIN3 mRNA is high.The expression of the two is negatively correlated.The high expression of PLIN3 mRNA is associated with poor prognosis in glioma patients.
