目的 探讨己糖激酶结构域成分 1(HKDC1)对人结直肠癌细胞增殖、迁移、侵袭能力的影响。方法 HT-29、SW480细胞进行瞬时转染,设置HKDC1-siRNA组、阴性对照组及空白组。同时收集 2020 年 3 月至 2022 年 5 月河北北方学院附属第一医院接受手术的 50 例患者的结直肠癌组织和癌旁正常组织。RT-qPCR实验检测HKDC1 mRNA表达;Western blot实验检测HKDC1 蛋白表达;CCK-8 实验、划痕实验和Transwell实验检测细胞增殖、迁移和侵袭能力。结果 HT-29 细胞HKDC1 mRNA的表达量高于SW480 细胞(P<0。05),故选取HT-29细胞株用于后续实验。结直肠癌组织中HKDC1 mRNA、光密度值高于癌旁正常组织(P<0。05)。空白组与阴性对照组HKDC1 mRNA、蛋白表达水平比较,差异无统计学意义(P>0。05);HKDC1-siRNA组HKDC1 mRNA、蛋白表达水平低于阴性对照组(P<0。05)。空白组与阴性对照组光密度值比较,差异无统计学意义(P>0。05);HKDC1-siRNA组光密度值低于阴性对照组(P<0。05)。空白组与阴性对照组细胞迁移率比较,差异无统计学意义(P>0。05);HKDC1-siRNA组细胞迁移率低于阴性对照组(P<0。05)。空白组与阴性对照组细胞迁移数目比较,差异无统计学意义(P>0。05);HKDC1-siRNA组细胞迁移数目低于阴性对照组(P<0。05)。结论 HKDC1在结直肠癌组织及细胞中高表达,可促进结直肠癌细胞的增殖、迁移及侵袭能力,有望成为结直肠癌基因治疗的新靶点。
Effect of Hexokinase domain component 1 gene knockout on the prolifer-ation,migration,and invasion ability of human colorectal cancer cells
Objective To investigate the effect of hexokinase domain component 1(HKDC1)on the proliferation,migration,and invasion of human colorectal cancer cells.Methods HT-29 and SW480 cells were transfected immediately,and HKDC1-siRNA group,negative control group,and blank group were set up.Meanwhile,colorectal cancer tissues and adjacent normal tissues of 50 patients who underwent surgery in the First Affiliated Hospital of Hebei North University from March 2020 to May 2022 were collected.The expression of HKDC1 mRNA was detected by RT-qPCR;the expression of HKDC1 protein was detected by Western blot;cell proliferation,migration,and invasion were detected by CCK-8 assay,scratch assay,and Transwell assay.Results The expression level of HKDC1 mRNA in HT-29 cell was higher than that in SW480 cell(P<0.05),therefore,HT-29 cell lines were selected for subsequent experiments.The expression levels of HKDC1 mRNA and optical density value in colorectal cancer tissues were higher than those in adjacent normal tissues(P<0.05).There were no significant differences in the expression levels of HKDC1 mRNA and protein between blank group and negative control group(P>0.05);the expression levels of HKDC1 mRNA and protein in HKDC1-siRNA group were lower than those in negative control group(P<0.05).There was no significant difference in optical density value between blank group and negative control group(P>0.05);the optical density value of HKDC1-siRNA group was lower than that of negative control group(P<0.05).There was no significant difference in cell migration rate between blank group and negative control group(P>0.05);the cell migration rate of HKDC1-siRNA group was lower than that of negative control group(P<0.05).There was no significant difference in the number of cell migration between blank group and negative control group(P>0.05);the number of cell migration in HKDC1-siRNA group was lower than that in negative control group(P<0.05).Conclusion HKDC1 is highly expressed in colorectal cancer tissues and cells,which can promote the proliferation,migration,and invasion ability of colorectal cancer cells,and is expected to become a new target for colorectal cancer gene therapy.
