铁死亡抑制剂在减轻放射性心脏病中的可能作用靶点及分子机制研究进展

摘要
放射性心脏病(RIHD)是放射治疗后影响患者生存质量和远期预后的重要原因之一,目前发病机制研究多聚焦于炎症反应、氧化应激和线粒体损伤等.铁死亡是一种以铁稳态破坏、脂质过氧化物生成为特点的新型调节性细胞死亡形式,发生过程中活性氧等成分的产生可能会加剧RIHD的进展,因此,铁死亡作为靶点应用于RIHD的治疗具有潜在意义.本文综述铁死亡和RIHD最新研究进展中相同的发病机制及分子通路,阐明铁死亡在RIHD中可能的作用靶点,为RIHD的预防及治疗提供新的思路.
Abstract
Radiation-induced heart disease(RIHD)is one of the important causes affecting quality of life and long-term prognosis of patients after radiotherapy.At present,pathogenesis research mainly focuses on inflammatory response,oxidative stress,and mitochondrial damage.Ferroptosis is a new form of regulatory cell death characterized by iron homeostasis destruction and lipid peroxidation.Production of reactive oxygen during the process may aggravate progress of RIHD.Therefore,ferroptosis as a target for the treatment of RIHD has potential significance.This article reviews the same pathogenesis and molecular pathways in the latest research progress of ferroptosis and RIHD,clarifies possible targets of ferroptosis in RI-HD,and provides a new idea for prevention and treatment of RIHD.
