Effects of ionizing radiation on expression of iron death driver SOCS1 and repressor FTH1 in nasopharyngeal carcinoma
Objective To explore the expression levels of iron death driver suppressors of cytokine signaling1(SOCS1)and repressor ferritin heavy chalin 1(FTH1)in nasopharyngeal carcinoma and the effects of ionizing radiation on them.Methods The expression levels of SOCS1 and FTH1 in the nasopharyngeal carcinoma datasets GSE53819(18 nasopharyngeal carcinoma samples and 18 nasopharyngeal normal tissue samples)and GSE12452(31 nasopharyngeal carcinoma samples and ten nasopharyngeal normal tissue samples)from the GEO database were analyzed,and the relationship between clinical characteristics such as different tumor stages and lymphatic metastasis status and primary-recurrent type and the levels of SOCS1 and FTH1 expression were analyzed in combination with GSE62328(30 nasopharyngeal carcinoma samples);the mRNA expression data in radioresistant nasopharyngeal carcinoma CNE2-IR cells and radiosensitive nasopharyngeal carcinoma CNE2 cells in the GSE48501 dataset were utilized to analyze the expression levels of SOCS1 versus FTH1 in the radioresistant and radiosensitive nasopharyngeal carcinoma cell lines;nasopharyngeal carcinoma C666-1 cells irradiated by 2 Gy of ionizing radiation were used as the experimental group,and nasopharyngeal carcinoma C666-1 cells irradiated by 0 Gy of ionizing radiation were used as the control group,and RT-qPCR was applied to detect the expression levels of SOCS1 and FTH1 in them,respectively.Results GEO database analysis showed that the expression levels of SOCS1 and FTH1 were higher in nasopharyngeal cancer tissues than those in normal nasopharyngeal tissues(P<0.05).Compared with nasopharyngeal cancer samples with lymph node metastasis,the expression levels of SOCS1 and FTH1 were higher in nasopharyngeal cancer samples without lymph node metastasis(P<0.05).There was not significant difference in the expression levels of SOCS1 and FTH1 in radioresistant and radiosensitive nasopharyngeal cancer cell lines(P>0.05).The results of real-time fluorescence quantitative PCR suggested that the expression level of SOCS1 in C666-1 cells of the experimental group was higher than that of the control group,and the expression level of FTH1 was lower than that of the control group(P<0.05).Conclusion Ferroptosis driver SOCS1 and suppressor FTH1 are differentially expressed in nasopharyngeal carcinoma and may serve as potential targets for nasopharyngeal carcinoma radiotherapy.
