Expression level and mechanism of Kindlin-3 and β2 integrin in patients with acute coronary syndrome
Objective To explore Kindlin-3 and β2 integrin expression levels in the peripheral blood of patients with acute coronary syndrome(ACS)and the associated mechanisms.Methods A total of 80 patients with ACS admitted to Xiangyang Central Hospital Affiliated to Hubei University of Arts and Sciences from September 2023 to January 2024 were selected,including 30 patients with unstable angina pectoris(UA)as UA group,26 patients with acute ST segment elevation myocar-dial infarction(STEMI),and 24 patients with acute non-ST segment elevation myocardial infarction(NSTEMI),the latter two were collectively known as the acute myocardial infarction(AMI)group.Additionally,twenty patients had normal coronary angiography and thirty patients had stable angina pectoris(SAP),forming the control.Flow cytometry was used to detect the ex-pression of β2 integrins(LFA-1 and Mac-1)in peripheral blood leukocytes.Enzyme linked immunsorbent assay was used to measure the levels of fractalkine,ICAM-1,C-reactive protein(CRP),and interleukin-6(IL-6)in venous serum.Western blot was used to determine the levels of Kindlin-3,FAK,and AKT proteins in peripheral blood leukocytes.Results Patients in the AMI group had higher levels of peripheral blood white blood cell count(WBC)and neutrophils count(NEUT)than those in the other three groups(P<0.05).The percentage of LFA-1 and Mac-1 positive cells in peripheral blood leuko-cytes of patients in the UA and AMI groups was higher than that of patients in the normal and SAP groups(P<0.05);the serum levels of Fractalkine,ICAM-1,and IL-6 were higher in patients in the UA group than those in the normal and SAP groups(P<0.05);the serum levels of Fractalkine,ICAM-1,CRP,and IL-6 were higher in patients in the AMI group than the other three groups(P<0.05).The levels of FAK proteins were higher in the UA group and AMI group than the normal and SAP groups(P<0.05);AKT protein levels were higher in the UA and AMI groups than the normal and SAP groups(P<0.05).The levels of FAK and AKT proteins were higher in the SAP group than the normal group(P<0.05).The expression levels of Kindlin-3 proteins were higher in the SAP group and UA group than the normal group(P<0.05),but decreased in the AMI group compared with the normal group(P<0.05).Conclusion Increased expression of Kindlin-3 and[3 2 integrins during ACS is accompanied by activation of the signalling pathway FAK-PI3K/AKT and release of inflammatory factors,suggesting their involvement in the inflammatory response to ACS.
