目的 基于Box-Behnken响应面法与反向传播(BP)神经网络优选千金子总蛋白最佳提取工艺.方法 以千金子总蛋白得率和纯度为指标,对碱提酸沉法、硫酸铵盐析法和超声提取法进行比较,确定千金子总蛋白提取方式;采用Box-Behnken响应面法结合BP神经网络优化千金子总蛋白提取工艺并进行验证;利用硫酸铵盐析法沉淀千金子总蛋白以确定不同硫酸铵饱和度盐析时对总蛋白含量的影响.结果 硫酸铵盐析法提取千金子总蛋白效果较好.采用Box-Behnken响应面法和BP神经网络优化得到的总蛋白提取率较高,最佳提取工艺为料液比为1:23(g/ml),提取时间为3.2 h,磷酸盐缓冲液浓度为0.02 mol/L,总蛋白提取率为19.87%,RSD值为0.30%.盐析用硫酸铵饱和度为90%时,总蛋自已基本沉淀完全.结论 基于Box-Behnken响应面法和BP神经网络优选的工艺稳定可行,模型预测效果较好,可为千金子总蛋白的提取工艺提供参考.
Study on the extraction process of total protein from Euphorbiae Semen based on Box-Behnken response surface methodology and back propaga-tion neural network
Objective To optimize the extraction process of total protein from Euphorbiae Semen based on Box-Behnken response surface methodology and back propagation(BP)neural network.Methods With the total protein extraction rate and purity as the index,the extraction method of total protein from Euphorbiae Semen was determined by comparing the method of alkali extraction and acid precipitation method,ammonium sulfate salting-out method,and ultrasonic extraction method.The Box-Behnken response surface methodology combined with BP neural network was used to optimize the extraction process of total protein and verify it.The total protein from Euphorbiae Semen was precipitated by ammonium sulfate salting-out method to determine the effect of different ammonium sulfate saturation salting-out on total protein content.Results The effect of ammonium sulfate salting-out method on the extraction of total protein from Euphorbiae Semen was better.The total protein extraction rate optimized by Box-Behnken response surface methodology and BP neural network was higher,and the optimal extraction process was as follows:the ratio of solid to liquid was 1:23(g/ml),the extraction time was 3.2 h,and the phosphate buffer solution concentration was 0.02 mol/L,and the extraction rate of total protein was 19.87%,RSD value was 0.30%.When the saturation of ammonium sulfate for salting-out was 90%,the total protein was basically precipitated completely.Conclusion The optimized process based on Box-Behnken response surface methodology and BP neural network is stable and feasible,and the model prediction effect is good,which can provide reference for the extraction process of Euphorbiae Semen.
