药用植物冷水花DNA条形码分析和基因组大小测定

摘要
目的:鉴定采集到的冷水花属植物样本并测定其基因组大小.方法:用聚合酶链式反应扩增样品的ITS2、rbcL和psbA-trnH序列并测序,通过比对Nr核酸数据库确定物种;以玉米Mo17 为对照,用流式细胞术测定其基因组大小.结果:本研究采集到的冷水花植物与冷水花(Pilea notata)ITS2 序列一致性为 100%,其基因组大小约为 498.4 Mb.结论:ITS2 序列可以较好地区分冷水花属植物,采用流式细胞术可以简便地估算待测植物样品的基因组大小.
Abstract
Objective To identify the collected Pilea L.plant sample and estimate its genome size.Methods PCR amplifica-tion and sequencing were performed for the ITS2,rbcL and psbA-trnH sequences,and then sequence alignments against the Nr database were performed for species identification.With the maize Mo17 genome as the control,flow cytometry was em-ployed to estimate the genome size of this plant.Results The ITS2 sequence of collected sample showed the identity of 100%to that of Pilea notata,with an estimated genome size of 498.4 Mb.Conclusion ITS2 sequences can distinguish dif-ferent species of Pilea and flow cytometry is a convenient way to estimate plant genome size.
