DNA Barcoding and Genome Size Estimation of the Medicinal Plant Pilea notata
Objective To identify the collected Pilea L.plant sample and estimate its genome size.Methods PCR amplifica-tion and sequencing were performed for the ITS2,rbcL and psbA-trnH sequences,and then sequence alignments against the Nr database were performed for species identification.With the maize Mo17 genome as the control,flow cytometry was em-ployed to estimate the genome size of this plant.Results The ITS2 sequence of collected sample showed the identity of 100%to that of Pilea notata,with an estimated genome size of 498.4 Mb.Conclusion ITS2 sequences can distinguish dif-ferent species of Pilea and flow cytometry is a convenient way to estimate plant genome size.
