目的:探讨蛋白磷酸酶PPM1F对非小细胞肺癌增殖和迁移的影响.方法:构建PPM1F敲低载体,通过Western blotting 验证PPM1F的敲低效率.通过CCK-8 实验、克隆形成实验、迁移实验、侵袭实验和流式细胞术检测PPM1F对非小细胞肺癌细胞功能的影响.结果:成功构建了PPM1F敲低质粒.敲低PPM1F抑制了非小细胞肺癌生长、增殖、迁移和侵袭能力.同时敲低 PPM1F,非小细胞肺癌细胞凋亡率增加,细胞在 G1 期累积.结论:PPM1F在非小细胞肺癌中以癌基因的作用发挥功能.
The Protein Phosphatase PPM1F Promotes the Progression of Non-Small Cell Lung Cancer:a Review
Objective To investigate the effects of the protein phosphatase PPM1F on the proliferation and metastasis of non-small cell lung cancer(NSCLC).Methods The vector for PPM1F knockdown was constructed,and Western blotting was employed to verify the knockdown efficiency of PPM1F.The effect of PPM1F on the function of NSCLC cells was deter-mined by the CCK-8 assay,colony formation assay,Transwell assay,invasion assay and flow cytometry.Results The plas-mid for PPM1F knockdown was successfully constructed.PPM1F knockdown inhibited the growth,proliferation,migration and invasion of NSCLC cells.When PPM1F was knocked down,the apoptosis rate of NSCLC cells increased,and the cells accumulated in the G1 phase.Conclusion PPM1F plays a role of oncogene in NSCLC.
