摘要

目的:优选毛连菜总有机酸提取工艺.方法:在单因素试验基础上,以液料比、提取时间及乙醇体积分数为自变量,以有机酸提取率为因变量,采用星点设计-响应面法优化毛连菜总有机酸提取工艺.结果:毛连菜总有机酸的最佳提取工艺为:液料比29 ∶ 1(mL·g-1)、乙醇体积分数67%、提取时间43 min.验证试验测得总有机酸提取率为(1.65±0.02)%,(n=3,RSD=0.92%),与模型预测值结果接近.结论:优化建立的提取工艺简单,工艺参数可靠,提取效果良好,为毛连菜总有机酸的后续研究奠定基础.

Abstract

Objective To optimize the extraction process of total organic acids from Picris hieracioides L..Methods Based on single factor test,the extraction process of total organic acids from Picris hieracioides L.was optimized by using central composite design-response surface method,with liquid-solid ratio,extraction time and ethanol volume fraction as inde-pendent variables,and extraction yield of total organic acids as dependent variable.Results The optimum ultrasonic extrac-tion process of total organic acids from Picris hieracioides L.was as follows:liquid material ratio 29 ∶ 1(mL·g-1),ethanol volume fraction 67%,and extraction time of 43 min.The verification experiment yielded a total organic acid extraction rate of(1.65±0.02)%(n=3,RSD=0.92%),which is close to the predicted value by the model.Conclusion The optimized extraction technology is simple,the process parameters are reliable,and the extraction effect is ideal,laying a foundation for the subsequent studies on total organic acids from Picris hieracioides L..
