Thin Endometrium Modeling in Rats with 95%Ethanol:An Experimental Study
Objective To explore the application value of three modeling methods for thin endometrium rats and their stabili-ty.Methods Three thin endometrium rat models were established using 95%ethanol,lipopolysaccharide(LPS),and me-chanical scraping separately.Endometrium tissue was collected one week after surgery.HE staining and Masson staining were performed to detect endometrium thickness and fibrosis area,and immunohistochemistry was used to evaluate the re-generation of endometrium cells by detecting cytokeratin 19(CK19)and vimentin.Mitochondrial apoptosis-inducing factor 2 was also analyzed to detect the mitochondrial apoptosis in the endometrium cells.Results All three methods were success-ful in thinning the endometrium of rats.However,under light microscopy,the structure of the endometrium in the mechani-cal scraping and LPS groups was found to be disordered and uneven,which made it difficult to uniformly compare the endo-metrium thickness.In comparison to the control group,the 95%ethanol group showed consistent and appropriate chemical damage,leading to thinned endometrium and increased fibrosis area.Immunohistochemical staining revealed significant re-ductions in CK19 and vimentin in the endometrium of the 95%ethanol group,while mitochondrial apoptosis-inducing fac-tor 2 was significantly increased.Conclusion Modeling with 95%ethanol was the most stable and efficient method for simu-lating the thin endometrium in rats,which can be utilized for subsequent scientific research.
