目的:采用Box-Behnken响应面法(RSM)优化娑罗子中七叶皂苷A的超声-微波协同提取工艺。方法:采用HPLC法测定七叶皂苷A的含量,单因素试验考察提取溶剂、料液比、超声功率、微波功率、提取温度、提取时间对七叶皂苷A提取率的影响,Box-Behnken响应面法优化超声-微波协同提取工艺。结果:优化所得最佳提取工艺为:提取溶剂40%乙醇(V/V),料液比1︰34(g/mL),超声功率230 W,微波功率200 W,提取温度40℃,提取时间10 min。在此工艺条件下对三批次药材进行验证,提取率为0。7625%,与模型预测值0。7587%接近,表明所建模型可靠有效。结论:超声-微波协同提取工艺提取时间短、提取率高、稳定可靠且绿色环保,可为工业上娑罗子中七叶皂苷A的提取提供参考。
Optimization of Ultrasound-Microwave Synergistic Extraction of Aescin A from Aesculi Semen
Objective To optimize the ultrasound-microwave synergitic extraction of Aescin A from Aesculi Semen using the Box-Behnken response surface methodology ( RSM) .Methods The content of Aescin A was determined by HPLC.Single factor tests were conducted to investigate the effects of extraction solvent,solid-liquid ratio,ultrasonic power,microwave power,extraction temperature,and extraction time on the extraction of Aescin A.RSM was used to optimize the ultrasound-microwave synergistic extraction process.Results The optimal extraction process was as follows:extraction solvent of 40%ethanol ( V/V),solid-liquid ratio at 1︰34 ( g/mL),ultrasonic power of 230 W,microwave power of 200 W,extraction temperature of 40℃,and extraction time of 10 min.Under this condition,the extraction rate of Aescin A from Semen Aes-culi of three batches was 0.7625%,which was close to the predicted 0.7587%,indicating the reliability and effectiveness of the model.Conclusion Ultrasound-microwave synergistic extraction has the advantages of short extraction time,high ex-traction rate,stability,reliability,and environmental protection,which provides a certain reference for the extraction of Aescin A from Aesculi Semen in industrial applications.
