Optimization of Ultrasound-Microwave Synergistic Extraction of Aescin A from Aesculi Semen
Objective To optimize the ultrasound-microwave synergitic extraction of Aescin A from Aesculi Semen using the Box-Behnken response surface methodology ( RSM) .Methods The content of Aescin A was determined by HPLC.Single factor tests were conducted to investigate the effects of extraction solvent,solid-liquid ratio,ultrasonic power,microwave power,extraction temperature,and extraction time on the extraction of Aescin A.RSM was used to optimize the ultrasound-microwave synergistic extraction process.Results The optimal extraction process was as follows:extraction solvent of 40%ethanol ( V/V),solid-liquid ratio at 1︰34 ( g/mL),ultrasonic power of 230 W,microwave power of 200 W,extraction temperature of 40℃,and extraction time of 10 min.Under this condition,the extraction rate of Aescin A from Semen Aes-culi of three batches was 0.7625%,which was close to the predicted 0.7587%,indicating the reliability and effectiveness of the model.Conclusion Ultrasound-microwave synergistic extraction has the advantages of short extraction time,high ex-traction rate,stability,reliability,and environmental protection,which provides a certain reference for the extraction of Aescin A from Aesculi Semen in industrial applications.
