Curcumin-Induced Oxidative Stress Against Melanoma by DNA Damage:An In Vitro Study
Objective To verify whether curcumin ( CUR)-induced oxidative stress promotes apoptosis and inhibit prolifera-tion in melanoma cells by inducing DNA damage.Methods The uptake of CUR by melanoma cells and its effect on cell via-bility were detected using CCK-8 and flow cytometry.The impacts of CUR on reactive oxygen species ( ROS) and DNA damage in melanoma cells were examined using Western blot and comet assay.Results CUR was taken up by melanoma cells,and it significantly reduced the cell viability with increasing concentration.Additionally,CUR remarkably increased the ROS level,leading to DNA damage,apoptosis,and thus proliferation blockade in melanoma cells.Conclusion This study indicates that CUR-induced oxidative stress has anti-melanoma effect by inducing DNA damage,providing a theoret-ical basis for the application of CUR in the treatment of melanoma.
