药用菊花种质资源遗传多样性的ISSR分析

目的 分析药用菊花遗传多样性,为其种质资源评价和合理开发利用提供分子佐证.方法 用ISSR分子标记对不同采集地的31份种源(菊花29份、野菊1份、菊花脑1份)进行遗传关系分析.结果 22个引物共扩增出182个条带,其中149条具有多态性,多态位点百分率为81.87%,有效等位基因数(Ne)为1.348 1,Nei's基因多样性指数(He)为0.219 1,Shannon信息指数(I)为0.345 1.聚类分析和主坐标分析结果 相近,将31份种源大致分为野菊和菊花脑、北方药用菊花、南方药用菊花3个类群.结论 ISSR分子标记适合于药用菊花的品种鉴定和遗传多样性分析.
ISSR Analysis for genetic diversity of Chrysanthemum morifolium
Objective To investigate the genetic diversity of Chrysanthemum morifolium and provide the evidence for evaluation and exploitation of C. morifolium germplasm.Methods The genetic diversities of 31 germplasm from different habitats were investigated with the technique of inter-simple sequence repeat(ISSR),which were 29 of C.morifolium,1 of C.indicum,and 1 of C.nankingense.Results Twenty-two primers were selected to produce highly reproducible ISSR bands.Among 182 amplified bands,149 showed polymorphism,the percentage of polymorphic bands(PPB)reached to 81.87%.Observed effective number of alleles(Ne),Nei's gene diversity index(HP),and Shannon information index(I) were 1.348 1,0.219 1,and 0.345 1,respectively.Conclusion ISSR Method is suitable for identification and genetic diversity analysis of C.morifolium.
