Consistency of three PD-L1 antibodies immunohistochemical staining in cervical squamous cell carcinomas
Objective To investigate the expression of PD-L1 in the cervical squamous cell carcinomas and the consistency of IHC detection of three different antibodies,including 22C3,SP263,and E1L3N.Methods The samples of 89 patients with recurrent or metastatic cervical squamous cell carcinoma were collected from PLA General Hospital.The samples were respectively stained with 22C3,SP263 and E1L3N,and combined positive score(CPS)and tumor cell proportion score(TPS)were used.The consistency of antibodies was statistically analyzed at different thresholds.Results When the positive threshold of CPS≥ 1,the positive rate of PD-L1 was 22C3(98.31%),E1L3N(97.75%)and SP263(93.90%).When CPS ≥10,the positive rates were E1L3N(76.40%),22C3(72.88%)and SP263(69.51%).When the TPS threshold ≥1%,the positive rates were E1L3N(95.51%),22C3(93.22%)and SP263(92.68%).When TPS ≥10%,the positive rates were E1L3N(65.17%),22C3(64.41%)and SP263(60.98%).When CPS ≥ 1 or 10,and TPS≥1%or 10%,E1L3N and 22C3,SP263 and 22C3,and SP263 and E1L3N showed good consistency(Kappa=0.66-0.98).Whether CPS or TPS was used,intra-class correlation coefficient tests showed a high degree of agreement among the three antibodies(ICC 0.987,0.984,respectively).Conclusion The three antibodies,E1L3N,SP263,and 22C3,can be used interchangeably,offering greater selectivity for clinical testing requirements.
