Abnormality of TOP2A gene and its exprsssion in anaplastic large cell lymphoma
Objective To investigate TOP2A gene copy number alterations and protein expression in anaplastic large cell lymphoma(ALCL),and to further analyze related clinical and pathological significance.Methods Fifty-eight cases of ALCL tissues were collected.Immunohistochemistry and fluorescence in situ hybridization(FISH)were used to detect the expression of TOP2A protein and the genetic abnormality of TOP2A,respectively and corresponding relationship was analyzed.Results Among the 58 ALCL patients,41 cases were positive for TOP2A protein.Among 50 cases of ALCL,26 cases had TOP2A gene amplification.TOP2A protein and TOP2A gene amplification were associated with Ann Arbor stage,IPI score and Ki-67 Index(P<0.05).Conclusion TOP2A is closely related to the occurrence and development of ALCL and the proliferation of tumor.Patients with positive expression of TOP2A protein and amplification of TOP2A gene have longer survival time.
