Timing and staining effect of 8% hydrochloric acid decalcification solution on bone marrow tissue
Objective To explore the staining effect of 8% hydrochloric acid decalcification solution on bone marrow tissue with different duration.Methods A total of 180 cases of bone marrow biopsy samples were collected,and the bone marrow tissues were treated with 8% hydrochloric acid decalcifier at 70 min,90 min and 110 min,respectively.The effects of HE staining,immunohistochemical staining(EnVision method staining TdT,CD56,MPO)and special staining(Gomori reticular fiber staining)on bone marrow tissues of different decalcification treatment periods were observed.Results Compared with 70 min and 110 min treated with 8% hydrochloric acid decalcification solution,the tissue structure of paraffin sections of bone marrow tissue remained more complete.After HE staining,the morphological structure was clear and distinguishable,the contrast was good,the immunohistochemical staining was accurate and the background was clear,and the staining thickness of mesh fibers was clearly distinguishable.Conclusion 8% hydrochloric acid decalcification solution has good effect on bone marrow tissue decalcification in 90 min.
